A New Sensitive Method for Detection of Viroids

Background and Aims: Viroids are smallest known plant pathogens and cause several economically significant diseases. Until recently, viroid detection relied mainly on biological tests and indexing. Today various diagnostic techniques such as nucleic acid hybridization, southern blot and reverse transcription coupled with polymerase chain reaction (RT-PCR) are being used for detection and diag...

Positively charged polymer brush-functionalized filter paper for DNA sequence determination following Dot blot hybridization employing a pyrrolidinyl peptide nucleic acid probe.

As inspired by the Dot blot analysis, a well known technique in molecular biology and genetics for detecting biomolecules, a new paper-based platform for colorimetric detection of specific DNA sequences employing peptide nucleic acid (PNA) as a probe has been developed. In this particular study, a pyrrolidinyl PNA bearing a conformationally rigid d-prolyl-2-aminocyclopentanecarboxylic acid back...

Comparison of genomic homologies in the coxsackievirus B group by use of cDNA:RNA dot-blot hybridization.

Radioactive cDNAs to coxsackievirus B (CB) genomic RNAs were used to probe genomic homologies between RNAs of the CB group serotypes. CB5 shared approximately equivalent homology with the other five CB serotypes. CB1, CB2, and CB3 hybridized preferentially to CB5 in turn, suggesting that these three have diverged in one direction from CB5. CB4 and CB6 were more related to each other than to the...

Simple and Accurate Detection of Vibrio Cholera Using Triplex Dot Blotting Assay

Cholera outbreak is more common in developing countries. The causative agent of the disease is Vibrio cholerae strains O1 and O139. Traditional diagnostic testing for Vibrio is not always reliable, because Vibrio can enter a viable but non cultivable state. Therefore, nucleic acid-based tests have emerged as a useful alternative to traditional enrichment testing.       In this investigation, a...

دورگه‌سازی در محل؛ اصول و کاربردها : مقاله مروری

In situ hybridization (ISH) is a method that uses labeled complementary single strand DNA or RNA to localize specific DNA or RNA sequences in an intact cell or in a fixed tissue section. The main steps of ISH consist of: probe selection, tissue or sample preparation, pre-hybridization treatment, hybridization and washing, detection and control procedure. Probe selection is one of the important ...
